Lupin was founded in 1968 by D.B. Gupta, who started the company with a focus on tuberculosis drugs at a time when TB was endemic in India and affordable treatment was a genuine public health need. The company's early specialisation in anti-infectives gave it manufacturing capabilities and regulatory experience in complex drugs that proved useful as it expanded into other therapy areas. Manju D. Gupta, his widow, has continued as a significant presence in the business, with their children Vinita Gupta, who serves as CEO, and Nilesh Gupta, Managing Director, running the operational side.

Lupin is now a significant global generics company with a strong position in the US market, where it competes in branded generics and complex formulations. The US business, built through product approvals and distribution relationships accumulated over two decades, generates a substantial portion of group revenues and operates at the high-regulation, high-stakes end of the generic pharmaceuticals market where manufacturing quality, intellectual property management and regulatory relationships are the decisive competitive variables.
